ABSTRACT

Objective To study the effects of apoptosis of the tumor cells in rabbit liver VX2 carcinoma after treatment by radiofrequency ablation(RFA) combined with trans arterial chemoembolization(TACE) and high‐frequency hyperthermia(HFH) . Methods Rabbit liver VX2 carcinoma model was established .Rabbit liver VX2 tumor models were divided into the following group:group A ,RFA+ TACE;group B ,RFA + TACE + HFH ;group C ,RFA + HFH ;group D ,TACE+ HFH .The changes of serum ALT was detected to realize the safety of the treatment .Cell apoptosis were detected by DNA agarose gel electrophoresis and terminal deoxynucleotidyltransferase‐mediated Dutp nick end‐labeling(TUNEL) assay ;SP immunohistochemistry ,Western blot and Real‐time quantitative PCR(RT‐PCR) were used to detect Caspase‐3 protein and mRNA expression levels .Results The changes of serum ALT in group B was significantly higher .Compared with other groups ,the apoptosis index in group B was increased marked‐ly(P<0 .05) .Western blot and RT‐PCR Caspase‐3 protein and mRNA levels in group B were higher than the other groups(P<0 .05) .Conclusion RFA+ TACE+ HFH can effectively kill tumor cells and promote apoptosis of tumor cells ,but ,at the same time ,can damage liver function .

ABSTRACT

Objective To investigate the relationship between polymorphisms of gene promoter region INS 5′UTR single nu-cleotide and type 2 diabetes and serum IAA-Ab levels.Methods By Sequenom MassArray SNP genotyping detection technology, INS 3 pyomter regime single nucleotide polymorphisms (rs689,rs714641 77 and rs3842738)of 497 patients in Chongqing with type 2 diabetes cases(treatment group)and 500 cases(control group)were genotyped and analyzed.IAA-Ab levels in diabetes patients was detected.Theχ2 test statistic was used to analyze the treatment group and control groups.The genotype frequency distribution of IAA-Ab-positive and negative groups SNP was analyzed by non-conditional logistic regression,adjusted for sex,age impact,cal-culated the odds ratio (OR)and 95 % confidence interval(CI ).The polymorphic loci with type 2 diabetes susceptibility and serum GAD-Ab levels was evaluated.Results The genotype frequency distribution of rs689AA,TT and AT was 58.75%,28.77% and 12.47%,respectively.The control group are 50.40%,35.60% and 14.00% respectively.The difference was statistically significant (χ2 =3.923,P <0.05).Compared with the genotype of AA,TT genotype can decrease risky of diabetes,with OR values 0.35(95%CI :0.18-1.06).There was significant difference of AA,TT,AT genotypes between IAA-Ab negative and IAA-Ab positive pa-tients (P < 0.05 ).Conclusion INS polymorphisms might be related to the risky of type 2 diabetes and serum IAA-Ab level in chinses population.

ABSTRACT

<p><b>OBJECTIVE</b>To study the changes in hypoxia-inducible factor (HIF1α, HIF2α) in the residual tumor cells in nude mice bearing hepatocellular carcinoma (HCC) following treatment with high-intensity focused ultrasound (HIFU).</p><p><b>METHODS</b>Thirty nude mice bearing human HCC received treatment with HIFU. At 1, 3, and 5 days and 1 and 2 weeks after the treatment, the mice were examined for pathological changes of the residual tumor with HE staining; SP immunohistochemistry, Western blotting and real-time quantitative PCR were used to detect the protein and mRNA expressions of HIF1α and HIF2α in the tumor.</p><p><b>RESULTS</b>HE staining revealed the presence of residual tumor cells and large necrotic areas after the treatment. Immunohistochemistry showed a gradual increment of HIF1α protein and mRNA expressions after the treatment, reaching the peak level at 3 days (P<0.05) followed by progressive reduction at 5 days and 1 and 2 weeks. HIF2α expressions at either the protein or mRNA levels exhibited no significant changes within 3 days after the treatment (P>0.05) but increased significantly at 5 days and 1 and 2 weeks (P<0.05).</p><p><b>CONCLUSION</b>The changes of HIF1α and HIF2α in the residual tumor after HIFU treatment in nude mice bearing HCC can be associated with tumor cell apoptosis and angiogenesis after the treatment.</p>

ABSTRACT

Objective To compare the current therapeutic effects between endovenous radiofrequency treatment and conventional surgery for lower limb chronic venous insufficiency (CVI) ,and summarize operative skills and clinical value of endovenous radiofreq uency treatment on CVI .Methods Data of 45 limbs of 41 patients treated by endovenous radiofrequency treatment (endovenous ra-diofrequencytreatmentgroup)and20limbsof20patientsbytraditionalsurgery(traditionalsurgerygroup)wereanalyzedandcom-pared in terms of operation time ,postoperative pain ,postoperative hospitalized duration ,complications ,and one-year recurrence rate .Results Endovenous radiofrequency treatment group had less postoperative pain and shorter postoperarive hospitalized dura-tion than traditional surgery group(P0 .05) . The two groups had few complications and had no recurrence in one year .Conclusion As a minimally invasive treatment for vari-cose veins ,endovenous radiofrequency treatment has less trauma ,rapid recovery ,safe and effective ,and it is worthy to be recommen-ded .

ABSTRACT

Objective: To evaluate the effectiveness that gelatin lines the injured vasuclar wall to prevent from restenosis.Methods: The animal model of restenosis was established by balloon dilation in femoral artery,and 30% gelatin was lined on injured vascular wall in experimental group.The samples were collected at the 2nd week and 4th week after operation.The neointimal area(NEA),neointimal area/media area(NEA/MA)and cell apoptosis were observed by image analysis system and TUNEL technique. Results: The intima of the control group was thicker than that of the experimental group,and more cells appeared in the control group.Compared with the control group,the NEA was reduced significantly in the experimental group at the 2nd and 4th week (0.67?0.25cm 2 vs 1.02?0.41cm 2,and 1.18?0.36cm 2 vs 1.46?0.15cm 2).The NEA/MA ratio in the experimental group was lower than the control group at the 2nd and 4th week(0.35?0.05% vs 0.54?0.14%,and 0.62?0.25% vs 0.75?0.10%).The apoptosis rate of vascular smooth muscle in the experimental group was higher than the control group at the 2nd and 4th week(18.2?5.8% vs 13.6?2.4%,and 15.1?2.1% vs 13.0?1.5%).These diffences were all significant (P

ABSTRACT

Objective To discuss the endothelial cell which was modified by exogenous anticoagulant genes contribute to the increase of antithrombosis activity of lined vascular prosthesis and the influence to other physiological functions of endothelial cells. Methods This summarized paper was made on literature review of recent years.Results The transfection of genes, including plasminogen activator (t-PA, u-PA, Urokinase), thrombomoduline (TM) and hirudin, etc, to endothelial cells resulted in not only the increase of antithrombosis activity of local vascular, but also the decrease of endothelial cell function in adherence and proliferation.Conclusion The increase of antithrombosis activity of lined vascular prosthesis has been done by exogenous genes. However, this technique ought to be studied, intensively.

ABSTRACT

Objective To explore the relationship among plasma cytokines’ level, adhesion molecules expression and skin damage in patients with chronic venous insufficiency (CVI) of lower extremities.Methods In 32 patients with CVI and 8 normal individuals as control, blood TNF ?, IL 1? and IL 2R were assayed with ELISA method; serum endothelial cell intercellular adhesion molecule 1(EC ICAM 1), polymorphonuclear CD 18 (PMN CD 18 ) and polymorphonuclear CD 11b (PMN CD 11b ) were assayed with immunohistochemical method; and ultrastructure of diseased veins was examined by electroscope.Results The results showed that the level of plasma TNF ? and IL 1? increased remarkably in Class 2-3 compared with Class 1 and control ( P 0.05) . The index of EC ICAM 1 and PMN CD 11b positively expression increased remarkably in Class 2-3 compared with that in Class 1 and control. ABSTRACT

Objective To investigate the irradiating effect of low intensive microwave(LIM) on pathological process of blood vessel restenosis(RS) and assess the probability of LIM irradiation to prevent was used RS.(Methods Forty-four) male healthy New Zealand rabbits were randomly divided into 2 groups.Fogarty catheter traumatize to the tunica intima of iliac artery so as to establish RS models.Two thousand four hundred and fifty MHz microwave with different power of 2,5 and 10 mW/cm~2 was used,locally to irradiate EIA in irradiating group(1 h/d).Specimens were obtained at different time of 3,7,14 and 28 d after operation.Morphological changes of tissues were observed with HE and EF staining and the area of tunica intima,tunica media and the rate of cavity stenosis were analyzed with image analysis system;apoptosis was detected with TUNEL;phenotype and microstructure of VSMC were observed with TEM.Results After microwave irradiating,inflammatory reaction in early period was suppressed,mural thrombus decreased,the proliferation and migration of VSMC depressed,the area of tunica intima and the rate of cavity stenosis obviously reduced comparing with the control group(P0.05).At other different time,however,the rate of apoptosis cells in irradiating groups obviously increased than that of the control group(P

ABSTRACT

Objective To study the relationship between cell apoptosis and vascular disease.Methods The paper reviewed the concept, character, gene control and signal transduction of apoptosis associated with vascular disease.Results Cytokine, radiation could induce apoptosis of endothelial cells, and caused vascular damage.The apoptosis of smooth muscle cells was related to the atherosclerosis and restenosis of vessels.Conclusion Cells apoptosis is related to vascular disease significantly.

ABSTRACT

AIM: To provide experimental evidence for gene therapy of thrombophilia disease, we constructed the eukaryotic expression plasmid with human thrombomodulin (hTM) gene and observed the alteration of hTM expression on the surface of human umbilical vein endothelial cells (HUVECs) with and without the reconstructive plasmid. METHODS: The whole expressive fragment of hTM gene was amplified by PCR from human genome. Both hTM gene and pcDNA3.1(+)/neo empty vector was digested by HindⅢ and EcoRⅠ. Two digested fragments were ligated into pcDNA3.1/hTM with T_4DNA ligase. After identifying, the reconstructive plasmid transfected into HUVECs using lipofectin. The hTM antigen on the HUVECs was detected by immunohistochemistry. RESULTS: The hTM reconstructive plasmid was confirmed by double endonuclease redigesting and sequencing. About 10% HUVECs were transfected by pcDNA3.1/hTM plasmid with lipofectin and the high-level hTM was detected on the transfected cells. CONCLUSION: We constructed the pcDNA3.1/hTM plasmid successfully, and it could be expressed on the HUVECs. [
